Idiocracy is a comedy, but it's got a hell of a lot of really amazing points in it. Check that out. I don't, personally, think that there are any movies out there which could actually "enlighten" you, based on how I define enlightenment anyway. There are a lot of them out there, though, they might get your mind really moving. For a few, go to www.freedocumentaries.org and watch "The Corporation", "Orwell Rolls in His Grave" and "Century of Self". I highly recommend all of those.
This movie is insanely good. It really puts some things in perspective and has a lot of great actors in it. In fact, I now want to rent this movie tonight and watch it again
Depends on what sort of enlightenment your seeking, no movie alone will do this, but it may guide on your path and provide some sort of stimulation for what your seeking. A waking life is good also A Scanner Darkly, not really the same kind of movie based off a book, but interesting all the same.
i've seen a lot of what's been recommended and to be honest most of it just bored/angered me. waking life was definitely worthwhile. if you want something "enlightening" eat some acid and watch happy feet lol. those animated films always seem to have a simple but important message to get across.

Dazed and Confused is one of my favorites. Keeps it simple but without being sophomoric and pointless. I also really like Donnie Darko. Waking Life is my favorite movie of all time, though. Woooo.
Dark City is one of my favorites. It deals with the human mind, questioning if our self is just a sum of all our memories or is there something else involved. Grave of the Fireflies is very good, and last years Into the Wild had some good insight. There's a lot of good movies out there with something to say, depends on what you like I guess. Different people will be "enlightened" by different things.
