Is there any advantage of going with a 400w HPS/MH over a 250w version if I plan on keeping the grow at 3 plants?

Yes, more watts, better coverage, penetration and yield.

-If the grow is taking place in a good sized room, are fans necessary?

Yes, for stem strength and fresh air.

-If the room lacks heating and the temp drops between 40-50ªF, willl growth be affected (keeping in mind the amount of heat that the lights produce)?

Yes, you dont want the temps to drop below 65-70 when the lights are on, or off.
